Science Magazine named lenacapavir its 2024 Breakthrough of the Year. In clinical trials, the twice-yearly injection provided complete protection against HIV infection — none of the 2,134 women who received the drug in the PURPOSE 1 trial contracted the virus.

The drug works through a novel mechanism, targeting multiple stages of the HIV life cycle simultaneously. Unlike daily pills, the long-acting injection only needs to be administered every six months, dramatically improving adherence.

In clinical trials, the twice-yearly injection provided complete protection against HIV infection — none of the 2,134 women who received the drug in the PURPOSE 1 trial contracted the virus.

"This could be transformative for HIV prevention," said Dr. Anthony Fauci. The breakthrough offers hope that the decades-long AIDS epidemic could finally be brought under control, especially in regions where daily medication regimens are challenging to maintain.
